The local side had to come from behind before getting the better of their opponents with the all-important goals scored by the McVeigh brothers, Paul and Brian.
It was another great result for the team managed by Shane Byrne. Last week, they beat Carlow IT 5-4 in the semi-final of the Colleges and Universities Football League.
DIT came to Letterkenny as the fancied team and they went ahead in the early stages of the second half when Aaron Brilly scored.
Letterkenny IT, who had strong appeals for a penalty turned down before the break, drew level soon after when this time they were awarded a penalty following a foul on Paul McVeigh.
The striker scored from the spot to make it 1-1.
Two minutes from the end, Brian McVeigh netted the winner, when he shot home from inside the area to send Letterkenny IT into the last four.
